Obviously there are many guides out there to aid designers in building usable websites but has anyone actually conducted usability testing before?

Never, even when I worked on an application packaged, marketed and distributed by a certain fortune 500 company (or companies). It is possible that they conducted a test and told us to make changes accordingly though.

Things like the Website Optimizer in Google make the process pretty easy now, would that encourage you to do it?

That's our specialty. There are a number of online validators, but all of them that I have seen make invalid assumptions. Take the advice they offer only as a starting point.

For example... the W3C WAI, Bobby, and Hermish validators all identify lack of a default script-type declaration as an issue, even though the W3C standard declares elsewhere that information sent as headers are acceptable substitutes. For example a PHP page sending the following header should be (and indeed acts as) a substitute parameter, yet fails the validators.

I wasn't thinking of usability from a disabled valid html point of view but rather determining which version of a web page design works best for your customers by using A/B or MVT testing.

I think an easy way to start is counting the number of clicks to accomplish each of the key objectives. You should aim for the least amount of clicks.

Second move links/buttons that support key objectives toward the top of the web page.

Third, remove any unnecessary / duplicate junk from the web page.

As a poor man test, you can always ask someone not familiar with your site to sit down for a few minutes and see what they do and/or having problem with.
